Detect endogenous peptides

Prevent protein fragments masking in vivo peptides

The presence of biologically-active peptides can be masked during analysis by the presence of protein fragments (peptides) derived from degradation processes that begin at the moment of sampling. Rapid heat stabilization prevents protein degradation thereby revealing endogenous peptides during subsequent analysis.

Using the Stabilizor system to heat-stabilize tissue samples:

  • Eliminates masking of in vivo expressed peptides

  • Prevents degradation during processing and analysis

  • Enables extraction of peptides in non-denaturing buffers without use of additives or precipitation steps

Detect more endogenous peptides

Results from a recent publication demonstrate an increased recovery of neuropeptides from tissue samples when utilizing heat stabilization followed by an optimal extraction protocol. (Neuropeptide profiling of the bovine hypothalamus: Thermal stabilization is an effective tool in inhibiting post-mortem degradation. Michelle L. Colgrave, et al., Proteomics 2011, 11, 1-13.)

Conventional protein identification criteria were applied when identifying neuropeptide precursors i.e. a minimum of two peptides derived from the same ‘protein’ identified with 95% confidence, hence any precursors containing only one peptide may be missed. At Sanofi-Aventis scientists detected more endogenous peptides in heat-stabilized mouse brain tissue compared to their standard protocol.
